Here Davide describes the development of a 384-well plate-based multi-parametric imaging assay designed to capture ex vivo primary islet proliferation, a known pathology related to Diabetes. The development of this assay allows for the positive identification of compounds that can selectively enhance islet beta-cell proliferation and reverse this known disease phenotype.

In this Special Issue of SLAS Discovery, the authors explore multiple aspects of cellular target engagement; from direct measurement of compound-target interaction using BRET technologies, through measurement of compound induced changes in protein stability and localisation, to in vivo drug distribution and target engagement studies. They also highlight technologies that enable a more holistic view of target engagement by mapping multiple compound-protein interactions in a single study. When applied in combination, these methods can build a strong platform of evidence linking compound to target to biological response, thereby impacting project attrition and medicines discovery success. 

Robert M. Campbell, Ph.D., is Senior Research Advisor at Eli Lilly and Company in Indianapolis, IN, where he heads the Cancer Cell Growth and Epigenetics Group. These articles are designed to provide a detailed, step-by-step guide to a particular experimental technique or methodology, enabling other researchers to replicate the work and build upon it.
